ABSTRACT Objective: To investigate the effect of ZnO nanocoating on mechanical properties of NiTi orthodontic wires and antibacterial activity. Methods: 0.016 x 0.022-in NiTi orthodontic wires were coated with ZnO nanoparticles using an electrochemical deposition method with three electrodes system in 0.1M Zn(NO3)2. Mechanical properties and frictional resistance of the coated wires were investigated using an universal testing machine. Antibacterial effect of ZnO coating was also investigated. Results: A stable adhered ZnO nanocoating on NiTi wires was obtained. The coated wires have a significant antibacterial activity against S. aureus, S. pyogens and E. coli, and a reduction of frictional forces by 34%. Conclusion: ZnO nanocoating may improve the antibacterial effects of NiTi wires and reduce the frictional resistance. Coating may be implanted in orthodontic practice for faster and safer treatment.


RESUMO Objetivos: Avaliar o efeito do nanorrevestimento de óxido de zinco (ZnO) sobre as propriedades mecânicas e propriedades antibacterianas de fios ortodônticos de NiTi. Métodos: Fios 0,016" x 0,022" de NiTi foram revestidos com nanopartículas de ZnO por meio de um método de deposição eletroquímica com um sistema de três eletrodos a 0,1M Zn(NO3)2. Uma máquina universal de testes foi utilizada para avaliar as propriedades mecânicas e a resistência friccional dos fios revestidos. Além disso, também foram analisadas as propriedades antibacterianas do revestimento de ZnO. Resultados: Obteve-se uma aderência estável das nanopartículas de ZnO sobre os fios NiTi. Os fios revestidos apresentaram atividade antibacteriana significativa contra S. aureus, S. pyogens e E. coli, e apresentaram uma redução de 34% na força de atrito. Conclusão: O revestimento com nanopartículas de óxido de zinco pode melhorar as propriedades antibacterianas e reduzir a resistência friccional dos fios de NiTi. Assim, o revestimento dos fios pode ser utilizado na Ortodontia visando tratamentos mais rápidos e seguros.

ABSTRACT

Abstract Calcium aluminate cement (CAC) has been highlighted as a promising alternative for endodontic use aiming at periapical tissue repair. However, its effects on dental pulp cells have been poorly explored. Objective: This study assessed the impact of calcium chloride (CaCl2) and bismuth oxide (Bi2O3) or zinc oxide (ZnO) additives on odontoblast cell response to CAC. Methodology: MDPC-23 cells were exposed for up to 14 d: 1) CAC with 2.8% CaCl2 and 25% ZnO (CACz); 2) CAC with 2.8% CaCl2 and 25% Bi2O3 (CACb); 3) CAC with 10% CaCl2 and 25% Bi2O3 (CACb+); or 4) mineral trioxide aggregate (MTA), placed on inserts. Non-exposed cultures served as control. Cell morphology, cell viability, gene expression of alkaline phosphatase (ALP), bone sialoprotein (BSP), and dentin matrix protein 1 (DMP-1), ALP activity, and extracellular matrix mineralization were evaluated. Data were compared using ANOVA (α=5%). Results: Lower cell density was detected only for MTA and CACb+ compared with Control, with areas showing reduced cell spreading. Cell viability was similar among groups at days one and three (p>0.05). CACb+ and MTA showed the lowest cell viability values at day seven (p>0.05). CACb and CACb+ promoted higher ALP and BSP expression compared with CACz (p<0.05); despite that, all cements supported ALP activity. Matrix mineralization were enhanced in CACb+ and MTA. Conclusion: In conclusion, CAC with Bi2O3, but not with ZnO, supported the expression of odontoblastic phenotype, but only the composition with 10% CaCl2 promoted mineralized matrix formation, rendering it suitable for dentin-pulp complex repair.

Abstract Although resin composites are widely used in the clinical practice, the development of recurrent caries at composite-tooth interface still remains as one of the principal shortcomings to be overcome in this field. Objectives To evaluate the activity against S. mutans biofilm of model resin composites incorporating different concentrations of ZnO-nanoparticles (ZnO-NP) and characterize their physicochemical properties. Materials and Methods Different concentrations of ZnO-NP (wt.%): E1=0, E2=0.5, E3=1, E4=2, E5=5 and E6=10 were incorporated into a model resin composite consisting of Bis-GMA-TEGDMA and barium borosilicate particles. The activity against S. mutans biofilm was evaluated by metabolic activity and lactic acid production. The following physicochemical properties were characterized: degree of conversion (DC%), flexural strength (FS), elastic modulus (EM), hardness (KHN), water sorption (Wsp), water solubility (Wsl) and translucency (TP). Results E3, E4, E5 and E6 decreased the biofilm metabolic activity and E5 and E6 decreased the lactic acid production (p<0.05). E6 presented the lowest DC% (p<0.05). No significant difference in FS and EM was found for all resin composites (p>0.05). E5 and E6 presented the lowest values of KHN (p<0.05). E6 presented a higher Wsp than E1 (p<0.05) and the highest Wsl (p<0.05). The translucency significantly decreased as the ZnO- NP concentration increased (p<0.05). Conclusions The incorporation of 2 - 5 wt.% of ZnO-NP could endow antibacterial activity to resin composites, without jeopardizing their physicochemical properties.

RESUMEN Objetivos Evaluar el efecto de las nanopartículas de ZnO, TiO2 y SiO2 sobre la viabilidad celular y la expresión génica de las interleuquinas 7 y 3 y del factor estimulante de colonias de granulocito - macrófago (GM-CSF) en Mus musculus. Materiales y métodos Se extrajo médula ósea roja de cinco roedores (Balb/c) para el estudio de viabilidad celular mediante la prueba de MTT. Por otro lado, grupos cinco roedores fueron inoculados vía intraperitoneal con dosis de 0,5; 1; 2,5; 5 y 10 mg/kg de nanopartículas de ZnO y SiO2 y de 5; 10; 15; 20 y 25 mg/kg de nanopartículas de TiO2, 30 h después, se obtuvo el ARN a partir de la médula ósea roja para los análisis de expresión génica empleando las técnicas de PCR y RT-PCR cuantitativa. Resultados Las nanopartículas de ZnO y SiO2 redujeron la viabilidad celular de una manera dosis-dependiente en un 37 y 26%, respectivamente, a partir de una dosis de 1 mg/kg. En cuanto al efecto sobre la expresión génica, a las dosis 5 y 10 mg/kg, las nanopartículas de TiO2 redujeron en mayor porcentaje la expresión de las interleuquinas 7 y 3 (55,3 y 70,2% respectivamente), con respecto a la expresión del GM-CSF, el mayor porcentaje de reducción lo produjo las nanopartículas de SiO2 (91%). Las nanopartículas de ZnO redujeron a partir de las dosis de 20 y 25 mg/kg. Conclusiones Las nanopartículas de ZnO, SiO2 y TiO2 alteran la viabilidad celular y la expresión génica en la médula ósea de ratón.


ABSTRACT Objectives To evaluate the effect of ZnO, TiO2 and SiO2 nanoparticles on cell viability and expression of the interleukin 7, interleukin 3, and granulocyte-macrophage colony stimulating factor (GM-CSF) genes in Mus musculus. Material and methods Red bone marrow was extracted from five Balb/c mice for the analysis of cell viability using the MTT test. The mice were divided into two groups of five each: one group was inoculated intraperitoneally with 0.5, 1.0, 2.5, 5.0, and 10 mg/kg of ZnO and SiO2 nanoparticles, respectively, and the other group was inoculated with 5.0, 10.0, 15.0, 20.0, and 25 mg/kg of TiO2 nanoparticles, respectively. Thirty hours later, RNA was extracted from the red bone marrow of the mice in both groups for gene expression analysis using quantitative PCR and RT-PCR. Results ZnO and SiO2 nanoparticles reduced cell viability in a dose-dependent manner by 37% and 26%, respectively, starting at a dose of 1 mg/kg. TiO2 nanoparticles at 5 mg/kg and 10 mg/kg reduced the gene expression of interleukins 7 and 3 by 55.3% and 70.2%, respectively, and SiO2 nanoparticles caused the greatest decrease (91%) in the expression of GM-CSF. ZnO nanoparticles reduced the expression of GM-CSF starting at doses of 20 mg/kg and 25 mg/kg. Conclusions ZnO, SiO2 and TiO2 nanoparticles affect cell viability and gene expression in the mouse bone marrow.

RESUMO: Objetivo: Estimar a prevalência de diagnóstico médico de asma na população adulta brasileira (≥ 18 anos). Métodos: Estudo transversal de base populacional com dados da Pesquisa Nacional de Saúde (PNS), de 2013; processo amostral por conglomerado com três estágios de seleção: setor censitário, domicílio e indivíduo. Calculou-se a prevalência e intervalo de confiança de 95% (IC95%) do desfecho "diagnóstico médico de asma" relatado pelo entrevistado e sua distribuição conforme variáveis demográficas, socioeconômicas, macrorregiões e zona urbana ou rural do país. Ainda foi investigado o manejo da asma naqueles que responderam afirmativamente sobre o diagnóstico médico; as análises foram ponderadas. Resultados: Foram entrevistados 60.202 adultos. A prevalência do diagnóstico médico de asma foi de 4,4% (IC95% 4,1 - 4,7), maior no sexo feminino, nos de cor branca, com maior escolaridade e moradores na região Sul; entre aqueles com diagnóstico médico, observou-se percentual elevado (38,2%) de crises de asma nos últimos 12 meses, com cerca de 80% usando medicação e 15% com limitação severa às atividades diárias. Conclusões: Apesar da estabilidade da prevalência da asma comparada a estudos anteriores no país, ainda são necessárias políticas para melhor manejo da doença.


ABSTRACT: Objective: To estimate the prevalence of asthma medical diagnosis among the adult Brazilian population (aged ≥ 18 years). Methods: This is a cross-sectional, population-based study from the 2013 National Health Survey (NHS); it is a sampling cluster process with three stages of selection: census tracts, households, and individuals. The prevalence and 95% confidence interval for the outcome "asthma medical diagnosis" reported by the interviewed subjects were calculated, besides its distribution according to demographic and socioeconomic variables, macroregions, and urban or rural area of the country. Management of the disease was also evaluated among those who reported asthma medical diagnosis and the analyses were weighted. Results: A total of 60,202 adults were interviewed. The prevalence of asthma medical diagnosis was 4.4% (95%CI 4.1 - 4.7), and it was higher among the female subjects, the white skin-colored subjects, those with higher educational level, and those who lived in the south of Brazil. Among those who reported asthma medical diagnosis, a high percentage of asthma attacks were seen in the last 12 months, with around 80% using medication and about 15% referring severe limitation to their daily activities. Conclusions: Although it seems there is asthma diagnosis stability in the country when compared with other researches, we still need public policies for improving the disease management.

ABSTRACT

This study evaluated the pH, calcium ion release and antimicrobial activity of EndoBinder (EB), containing different radiopacifiers: bismuth oxide (Bi2O3), zinc oxide (ZnO) or zirconium oxide (ZrO2), in comparison to MTA. For pH and calcium ion release tests, 5 specimens per group (n = 5) were immersed into 10 mL of distilled and deionized water at 37°C. After 2, 4, 12, 24, 48 h; 7, 14 and 28 days, the pH was measured and calcium ion release quantified in an atomic absorption spectrophotometer. For antimicrobial activity, the cements were tested against S. aureus, E. coli, E. faecalis and C. albicans, in triplicate. MTA presented higher values for pH and calcium ion release than the other groups, however, with no statistically significant difference after 28 days (p > 0.05); and the largest inhibition halos for all strains, with no significant difference (E. coli and E. faecalis) for pure EB and EB + Bi2O3 (p > 0.05). EB presented similar performance to that of MTA as regards pH and calcium ion release; however, when ZnO and ZrO2 were used, EB did not present antimicrobial activity against some strains.

ABSTRACT

This study aimed to evaluate by the intra-osseous implant technique the most commonly used materials for pulp therapy in pediatric dentistry: calcium hydroxide (CH), Guedes Pinto paste and CTZ paste, according to FDI (1980) and ANSI/ADA (1982) recommendations. Thirty guinea pigs, 10 for each material, divided into experimental periods of 4 and 12 weeks received one implant on each side of the lower jaw symphysis. The external lateral tube wall served as control for the technique. At the end of the observation periods, the animals were euthanized and specimens were prepared for routine histological examination. It was observed that CH and CTZ paste induced severe inflammation, a large amount of necrotic tissue, lymphocytes, foreign body cells and bone resorption, while Guedes Pinto Paste induced little or no inflammation in the 4-week observation period. After 12 weeks, the reactions to CH and Guedes Pinto paste were also absent/mild, presenting a general pattern of replacement by recently formed bone tissue while a moderate to severe inflammatory response was observed with CTZ paste. Guedes Pinto paste presented acceptable biocompatibility levels in both analyzed periods; CH only showed acceptable biocompatibility in the 12-week period while CTZ paste showed no biocompatibility in both periods. Among the tested materials, only Guedes Pinto paste presented an acceptable biocompatibility.


A pesquisa teve como objetivo avaliar a biocompatibilidade através da técnica de implantes intra-ósseos dos materiais utilizados em odontopediatria para tratamento pulpar: hidróxido de cálcio, pastas Guedes Pinto e CTZ, de acordo com as recomendações da FDI (1980) e ANSI/ADA(1982). Trinta guinea pigs, dez para cada material, divididos em períodos experimentais de 4 e 12 semanas receberam um implante em cada lado da sínfise mandibular. A parede lateral externa do copo serviu como controle para a técnica. No final dos períodos experimentais, os animais foram sacrificados e os espécimes preparados para o exame histológico de rotina. Observou-se que o hidróxido de cálcio e a pasta CTZ mostraram reação inflamatória severa, grande quantidade de tecido necrosado, linfócitos, células de corpo estranho e reabsorção óssea; enquanto a pasta Guedes Pinto induziu pouca ou nenhuma inflamação no período de 4 semanas. Após 12 semanas as reações para o hidróxido de cálcio e pasta Guedes Pinto foram ausentes/suaves apresentando um padrão geral de substituição por tecido ósseo neoformado, enquanto uma resposta inflamatória de moderada a severa foi observada para a pasta CTZ. A pasta Guedes Pinto apresentou níveis aceitáveis de biocompatibilidade nos dois períodos analisados; hidróxido de cálcio apresentou biocompatibilidade aceitável somente no período de 12 semanas e a pasta CTZ não mostrou biocompatibilidade em ambos os períodos. Entre estes, apenas a pasta Guedes Pinto apresentou níveis de biocompatibilidade nos dois períodos analisados.

This study evaluated the response of the subcutaneous connective tissue of BALB/c mice to root filling materials indicated for primary teeth: zinc oxide/eugenol cement (ZOE), Calen paste thickened with zinc oxide (Calen/ZO) and Sealapex sealer. The mice (n=102) received polyethylene tube implants with the materials, thereby forming 11 groups, as follows: I, II, III: Calen/ZO for 7, 21 and 63 days, respectively; IV, V, VI: Sealapex for 7, 21 and 63 days, respectively; VII, VIII, IX: ZOE for 7, 21 and 63 days, respectively; X and XI: empty tube for 7 and 21 days, respectively. The biopsied tissues were submitted to histological analysis (descriptive analysis and semi-quantitative analysis using a scoring system for collagen fiber formation, tissue thickness and inflammatory infiltrate). A quantitative analysis was performed by measuring the area and thickness of the granulomatous reactionary tissue (GRT). Data were analyzed by Kruskal-Wallis, ANOVA and Tukey's post-hoc tests (?=0.05). There was no significant difference (p>0.05) among the materials with respect to collagen fiber formation or GRT thickness. However, Calen/ZO produced the least severe inflammatory infiltrate (p<0.05). The area of the GRT was significantly smaller (p<0.05) for Calen/ZO and Sealapex. In conclusion, Calen/ZO presented the best tissue reaction, followed by Sealapex and ZOE.


Neste estudo avaliou-se a resposta do tecido conjuntivo subcutâneo de camundongos BALB/c a materiais obturadores de canal radicular de dentes decíduos: óxido de zinco/eugenol (OZE), pasta Calen® espessada com óxido de zinco (Calen/OZ) e cimento Sealapex®. Os camundongos (n=102) receberam implantes de tubos de polietileno??? e foram divididos em grupos: I, II, III - Calen/OZ (7, 21 e 63 dias, respectivamente); IV, V, VI - Sealapex (7, 21 e 63 dias, respectivamente); VII, VIII, IX - OZE (7, 21 e 63 dias, respectivamente); X, XI - tubo vazio (7 e 21 dias, respectivamente). Os tecidos foram submetidos ao processamento e análise histopatológica descritiva e por meio de escores do fibrosamento, espessura tecidual e infiltrado inflamatório. Para a análise quantitativa mensurou-se a área e a espessura do tecido granulomatoso reacional (TGR). Os resultados foram analisados pelos testes de Kruskal-Wallis, ANOVA e pós-teste de Tukey (?=0,05). Não houve diferença significante (p>0,05) entre os materiais, com relação ao fibrosamento e à espessura do TGR. Contudo, Calen/OZ apresentou infiltrado inflamatório de menor intensidade (p<0,05). A área do TGR foi menor (p<0,05) para Calen/OZ e Sealapex. Pôde-se concluir que Calen/OZ foi o material que apresentou a melhor compatibilidade tecidual, seguido pelos cimentos Sealapex e OZE.

ABSTRACT

The aim of this study was to evaluate the biocompatibility of sealers used in apical surgery in rat subcutaneous tissue. Sterile polyethylene tubes were filled with the following sealers: Sealapex, Sealapex with addition of zinc oxide, Sealer 26, Sealer 26 with thicker consistency (greater powder-to-resin ratio) and White MTA. The tubes were implanted in the dorsum of male rats and after 7, 21 and 42 days, the animals were killed, obtaining 5 specimens for each sealer in each evaluation period. The lateral surface of the tube was used as negative control. The inflammatory reaction to contact with the sealers was classified as absent, mild, moderate and severe. At 7 days, all sealers caused similar inflammatory reactions in the connective tissue of the animals, with most specimens presenting a moderate to intense chronic inflammatory reaction, with presence of multinucleated giant cells. At 21 days, Sealer 26 and Sealer 26 with thicker consistency presented more intense inflammatory reaction (p=0.004), whereas after 42 days, the inflammatory reaction ranged from absent to mild with statistically similar results for both materials (p=0.08). Except for MTA, all sealers presented foreign-body granulomatous reaction at 42 days. All sealers but Sealapex presented a statistically significant decrease of the inflammatory reaction over time. In conclusion, all sealers caused moderate to severe inflammation in the earlier evaluation period. However, Sealer 26 and Sealer 26 with thicker consistency caused more intense inflammatory reactions after 21 days of contact with the tissues and no granulomatous reaction was observed for MTA at the final period of analysis.


O objetivo deste estudo foi avaliar a biocompatibilidade de cimentos utilizados em cirurgia apical, em tecidos subcutâneos de ratos. Tubos de polietileno esterilizados foram preenchidos com os seguintes cimentos: Sealapex, Sealapex acrescido de óxido de zinco, Sealer 26, Sealer 26 espessado (maior proporção pó:resina) e MTA branco. Os tubos foram implantados no dorso de ratos machos e após 7, 21 e 42 dias, os animais foram mortos, obtendo 5 amostras por cimento em cada período analisado. A superfície lateral do tubo foi utilizada como controle negativo. A reação inflamatória em contato com os cimentos foram classificadas como ausente, leve, moderada e severa. Aos 7 dias, todos os cimentos induziram reações inflamatórias similares no tecido conjuntivo dos animais, com a maioria dos espécimes apresentando reação inflamatória crônica de moderada a intensa, com presença de células gigantes multinucleadas. Aos 21 dias, Sealer 26 e Sealer 26 espessado apresentaram reação inflamatória mais intensa (p=0,004), enquanto após 42 dias, a reação inflamatória variou de ausente a leve, com resultados estatisticamente semelhantes para ambos materiais (p=0,08). Com exceção do grupo MTA, todos os cimentos apresentavam reação granulomatosa de corpo estranho após 42 dias. Todos os grupos, exceto o Sealapex, apresentaram redução estatisticamente significante dos índices inflamatórios ao longo do tempo. Conclui-se que todos os cimentos induziram reação inflamatória de moderada a intensa no período inicial de análise. Entretanto, Sealer 26 e Sealer 26 espessado apresentaram reação inflamatória mais intensa após 21 dias de contato com os tecidos e reação granulomatosa não foi observada no grupo MTA no período final de análise.

ABSTRACT

The purpose of this study was to investigate the influence of serum and necrotic soft tissue on the antimicrobial activity of intracanal medicaments. The medicaments tested were: calcium hydroxyde/glycerin paste, calcium hydroxide/chlorhexidine paste, calcium hydroxide/camphorated paramonochlorophenol/glycerin paste, and chlorhexidine/zinc oxide paste. Survival of Enterococcus faecalis and Candida albicans exposed to the medicaments tested in the presence or absence of serum or necrotic tissue was monitored in three in vitro experiments where samples for culturing were taken at different time periods. The overall results demonstrated that the antimicrobial activity of all intracanal medicaments tested was slowed down in the presence of necrotic tissue. Calcium hydroxide pastes in glycerin or chlorhexidine were significantly affected by serum. Of the medicaments tested in this study, the least affected was the calcium hydroxide/camphorated paramonochlorophenol/glycerin paste.


O objetivo deste estudo foi avaliar a influência do soro e de tecido mole necrosado na atividade antimicrobiana de medicamentos intra-canais. Os medicamentos testados foram pastas de hidróxido de cálcio/glicerina, hidróxido de cálcio/clorexidina, hidróxido de cálcio/paramonoclorofenol canforado/glicerina e clorexidina/óxido de zinco. A sobrevivência de Enterococcus faecalis e Candida albicans expostos aos medicamentos na presença ou ausência de soro ou tecido necrosado foi monitorada em três experimentos in vitro nos quais amostras para cultura foram avaliadas em diferentes períodos de tempo. No geral, os resultados demonstraram que a atividade antimicrobiana de todos os medicamentos testados foi retardada na presença de soro ou de tecido necrosado. As pastas de hidróxido de cálcio em glicerina ou clorexidina foram significativamente afetadas pelo soro. Dos medicamentos testados, o menos afetado foi a pasta de hidróxido de cálcio/paramonoclorofenol canforado/glicerina.

ABSTRACT

This study evaluated in vitro the antibacterial activity of 4 root canal filling materials for primary teeth - zinc oxide and eugenol cement (ZOE), Calen paste thickened with zinc oxide (Calen/ZO), Sealapex sealer and EndoREZ sealer - against 5 bacterial strains commonly found in endodontic infections (Kocuria rhizophila, Enterococcus faecalis, Streptococcus mutans, Escherichia coli and Staphylococcus aureus) using the agar diffusion test (agar-well technique). Calen paste, 1 percent chlorhexidine digluconate (CHX) and distilled water served as controls. Seven wells per dish were made at equidistant points and immediately filled with the test and control materials. After incubation of the plates at 37oC for 24 h, the diameter of the zones of bacterial growth inhibition produced around the wells was measured (in mm) with a digital caliper under reflected light. Data were analyzed statistically by analysis of variance and Tukey's post-hoc test (?=0.05). There were statistically significant differences (p<0.0001) among the zones of bacterial growth inhibition produced by the different materials against all target microorganisms. K. rhizophila was inhibited more effectively (p<0.05) by ZOE, while Calen/ZO had its highest antibacterial activity against E. faecalis (p<0.05). S. mutans was inhibited by Calen/ZO, Sealapex and ZOE in the same intensity (p>0.05). E. coli was inhibited more effectively (p<0.05) by ZOE, followed by Calen/ZO and Sealapex. Calen/ZO and ZOE were equally effective (p>0.05) against S. aureus, while Sealapex had the lowest antibacterial efficacy (p<0.05) against this microorganism. EndoREZ presented antibacterial activity only against K. rhizophila and S. aureus. The Calen paste and Calen/ZO produced larger zones of inhibition than 1 percent CHX when the marker microorganism was E faecalis. Este estudo avaliou in vitro a atividade antibacteriana de 4 materiais obturadores de canais radiculares de dentes decíduos - cimento de óxido de zinco e eugenol (OZE), pasta Calen espessada com óxido de zinco (Calen/OZ), cimento Sealapex e cimento EndoREZ - sobre 5 cepas bacterianas comumente encontradas em infecções endodônticas: Kocuria rhizophila, Enterococcus faecalis, Streptococcus mutans, Escherichia coli e Staphylococcus aureus, usando o teste de difusão em ágar (técnica do poço). A pasta Calen, digluconato de clorexidina a 1 por cento (CHX) e água destilada foram usados como controle. Sete poços por placa foram preparados em pontos eqüidistantes e imediatamente preenchidos com os materiais experimentais e controle. Após incubação das placas a 37oC por 24 h, o diâmetro dos halos de inibição do crescimento bacteriano formados ao redor dos poços foi medido (em mm) com um paquímetro digital sob luz refletida. Os dados obtidos foram submetidos à análise de variância e ao pós-teste de Tukey (?=0,05). Com relação à atividade antibacteriana, evidenciaram-se diferenças estatisticamente significantes (p<0,0001) entre os halos de inibição formados pelos diferentes materiais, para todos os microrganismos avaliados. A K. rhizophila foi inibida mais eficazmente pelo OZE (p<0,05), enquanto que o E. faecalis foi inibido mais eficazmente pela Calen/OZ (p<0,05). O S. mutans foi inibido pela Calen/OZ, cimento Sealapex e OZE na mesma intensidade (p>0,05). A E. coli foi inibida mais eficazmente pelo OZE, seguido pela Calen/OZ e pelo cimento Sealapex (p<0,05). O S. aureus foi inibido pela Calen/OZ e OZE na mesma intensidade (p>0,05), e menos intensamente pelo cimento Sealapex (p<0,05). ABSTRACT

An important requirement for endodontic paste with antibiotics placed in direct contact with living tissues is biocompatibility. The aim of this study was to evaluate the paste biocompatibility prepared with zinc oxide (1.25mg), tetracycline (8mg) and thiamphenicol (26.67mg). The paste and its components were implanted separately through polyethylene sterile tubes of 10mm in length and 1.3mm in diameter, in the subcutaneous tissue of rats with the experiment control at intervals of 3, 7, 15 and 30 days. Each day 6 rats were used, being 3 of them with implant of the substances in four sites placed on the back of the animals and 3 sham animals where it was implanted the polyethylene empty tubes. The experimental animals were anesthetized in an intra-peritoneal way with ketamina and xilazina (0.75ml / g body weight). After the experimental periods, the animals were anesthetized with the same anesthetic overdose. It was held an excision biopsy of the implant area with 10 mm to the security limit included in paraffin following a plan of random histological cut and uniformlyisotropic or oriented cuts according to stereological principles, getting a statistical estimative of the relative amount of inflammatory cells or not on the test system, getting as a result the paste biocompatibility, being the zinc oxide the most toxic element for the cell quality found.


Un requisito importante para la pasta endodóntica preparada con antibióticos, que es colocada en contacto directo con los tejidos vivos es la biocompatibilidad. El objetivo de este estudio fue evaluar la biocompatibilidad de la pasta preparada con óxido del zinc (1,25mg), tetraciclina (8mg) y el tiamfenicol (26,67mg). La pasta y sus componentes fueron implantados por separado a través de tubos estériles de polietileno de 10 mm de longitud y de 1,3mm de diámetro en el tejido subcutáneo de ratas en intervalos de 3, 7, 15 y 30 días. Cada día, 6 ratas fueron implantadas en cuatro sitios ubicados en la parte posterior de los animales, 3 de ellas con el implante de las sustancias y 3 fueron implantados con los tubos de polietileno vacíos. Los animales del experimento fueron anestesiados intraperitteonealmen, con ketamina y xilasina (0,75 ml/g peso corporal). Después de los periodos experimentales, los animales fueron anestesiados con la misma sobredosis anestésica. Fue realizada una biopsia exisional del área del implante con 10 mm de límite de seguridad, luego se realizaron cortes histológico al azar uniformemente isotrópicos o orientados según los principios esteriológicos, consiguiendo un estimativo estadístico de la cantidad relativa de células inflamatorias en el sistema de prueba. Se obtuvieron resultados de la biocompatibilidad de la pasta, siendo el óxido del zinc el elemento mas tóxico según la cualidad de las células que fueron encontradas.

ABSTRACT

The purposes of this study were to evaluate the effectiveness of 2 percent chlorhexidine (CHX) gluconate gel, calcium hydroxide [Ca(OH)2] and their combination with iodoform and zinc oxide powder as intracanal medications against select microorganisms, and to measure the pH changes caused by these medications. Antimicrobial activity was determined by the agar diffusion method. The zones of growth inhibition were measured and the results were analyzed statistically by Kruskal-Wallis test (p<0.05). The pH of the pastes was measured right after preparation, after 24 h and 1 week later. The largest mean zones of microbial inhibition were produced by 2 percent CHX gel, followed by Ca(OH)2 + 2 percent CHX gel + iodoform, Ca(OH)2 + 2 percent CHX gel, Ca(OH)2 + 2 percent CHX gel + zinc oxide, and Ca(OH)2 + water. The mean pH of all medications stayed above 12.0 during the whole experiment, except for CHX gel (pH=7.0). The results of this study showed that all medications had antimicrobial activity, but the most effective against the tested microorganisms were 2 percent CHX gel, followed by its combination with Ca(OH)2 and iodoform.


O objetivo do estudo foi avaliar, in vitro, a efetividade antimicrobiana da clorexidina gel 2 por cento (CHX) e hidróxido de cálcio, isoladamente e associados com iodofórmio e pó de óxido de zinco como medicamentos intracanais frente a microrganismos e medidos pHs das diferentes medicações. A atividade antimicrobiana foi determinada pelo método de difusão em ágar. As áreas de inibição de crescimento foram medidas e os resultados estatisticamente analisados utilizando-se o teste de Kruskal-Wallis (p<0,05). O pH das pastas foi mensurado após a manipulação, após 24 h e após uma semana. Os resultados mostraram que a maior zona de inibição foi da CHX gel 2 por cento, seguida pelo Ca(OH)2 + 2 por cento CHX gel, Ca(OH)2 + 2 por cento CHX gel + iodofórmio, Ca(OH)2 + 2 por cento CHX gel +óxido de zinco, Ca(OH)2 + água. A média de pH de todos os medicamentos intracanais foi de 12 durante todo o experimento, exceto com CHX gel 2 por cento (pH=7,0). Estes resultados permitiram concluir que todos os medicamentos tiveram atividade antimicrobiana, no entanto, a maior foi da CHX gel 2 por cento, seguido da associação com o Ca(OH)2. e iodofórmio.

ABSTRACT

Foi propósito deste trabalho observar o processo de reparo de dentes de cães após obturação dos canais com dois cimentos diferentes, fazendo ou não a patência apical. Após uma sobreinstrumentação, os canais receberam um curativo de uma solução de corticosteróide-antibiótico por 7 dias, com o objetivo de obter invaginação de tecido conjuntivo para dentro dos canais. Após esse período, esse tecido foi totalmente removido em metade dos casos (grupos com patência apical) e preservados no restante dos casos (grupos sem patência apical). Os canais foram obturados pela técnica da condensação lateral empregando um cimento a base de hidróxido de cálcio (Sealer Plus) ou um cimento de Grossman (Fill Canal). Os animais foram sacrificados por overdose anestésica 60 dias após o tratamento endodôntico e as peças anatômicas foram obtidas e preparadas para análise histológica. Os dados obtidos foram analisados com base em diversos parâmetros histomorfológicos. Os resultados foram melhores nos grupos sem patência apical (p=0,01) do que nos grupos com patência. Dentre os cimentos estudados, os melhores resultados foram observados com o cimento Sealer Plus (p=0,01). Em conclusão, tanto a patência apical (presença ou ausência) quanto o tipo de material obturador de canal influíram no processo de reparo apical de dentes de cães com polpas vitais após tratamento endodôntico. O emprego de um cimento a base de hidróxido de cálcio em dentes sem patência apical promoveu os melhores resultados, dentre as condições experimentais propostas.

ABSTRACT

Las úlceras de miembros inferiores por insuficiencia venosa son una patología frecuente en pacientes por sobre la cuarta década de la vida, incapacitante, refractaria a la terapia y causal de hospitalización prolongada. La Pasta de Unna ha sido uno de los tratamientos más utilizados desde los últimos 100 años. Mostramos en este trabajo los beneficios del uso de un esquema secuencial de la cicatrización completa o preparación para un ulterior injerto de dichas úlceras; esta experiencia se fundamenta en otras investigaciones publicadas con anterioridad. Realizamos un estudio prospectivo en 23 pacientes, de ambos sexos, adultos, con úlceras de miembros inferiores, de diferentes etiologías, durante el periodo comprendido entre enero de 1986 hasta octubre de 1998. Se utilizaron diversos productos cicatrizantes y antibacterianos previos a la aplicación de la Pasta de Unna Modificada, además de un método práctico de utilización de ésta. Del total de 23 pacientes estudiados, la úlcera más frecuente fue por insuficiencia venosa en 21 pacientes (91,30 por ciento). Se excluyeron 6 pacientes por presentar: vasculopatía periférica no venosa, reinfección, evolución tórpida del injerto o retiro de la consulta. Se incluyen 17 pacientes susceptibles de evaluar. La respuesta con cicatrización fue buena en 14 pacientes (82,35 por ciento). Nuestra experiencia demuestra que el reposo, limpieza y antibioticoterapia tópica y/o sistémica previa y el recambio semanal de la bota hasta lograr su epitelización o apta para colocar injerto dermoepidérmico son necesarios. Es un método efectivo para el tratamiento de las úlceras por insuficiencia venosa

ABSTRACT

Four materials viz. zinc oxide-eugenol, iodoform paste, Kri paste, Maisto's paste and Vitapex (Calcium hydroxide + iodoform) were tested for their antibacterial effect against the aerobic and anerobic bacteria, viz. Staphylococcus aureus, Streptococcus viridans, Streptococcus faecalis, Bacteroides melaninogenicus and mixed bacterial culture; obtained from infected non-vital deciduous anterior teeth. The antimicrobial sensitivity was checked on BHI-agar plates using well method. The results showed that all the 4 materials were distinctly different from each other in their antimicrobial activity. Maisto's paste was invariably the superior most in its antibacterial efficacy (in comparison to all the 5 micro-organism strains). Iodoformized zincoxide eugenol appeared to be the second best followed by Kri paste. Vitapex showed the least antibacterial activity.

Estudos têm mostrado discreta reaçäo do tecido pulpar quando este é capeado com sistemas adesivos. Muitos pesquisadores têm relatado formaçäo de ponte de dentina adjacente à exposiçäo pulpar. Este estudo foi realizado para observar a capacidade de reparaçäo e formação de ponte de dentina de polpas mecanicamente expostas que foram capeadas com o sistema adesivo Scotchbond MP (Sbmp - grupo experimental) e com o cimento de óxido de zinco e eugenol (OZE - grupo controle). Em 24 ratos, 48 cavidades de Classe I ocolusal com exposiçäo da polpa foram realizadas, das quais 24 foram capeadas com Sbmp e o restante com OZE. As cavidades foram restauradas com a resina Z100. As maxilas foram removidas após intervalos de 7, 15, 30 e 60 dias. Cortes seriados dos dentes foram corados com hematoxilina e eosina e Brown e Brenn. Para o grupo experimental, aos 7 dias houve necrose do corno pulpar adjacente ao material. Com o decorrer dos períodos, o tecido pulpar näo apresentou capacidade de reorganizaçäo e formação de ponte de dentina. Aos 60 dias a polpa estava totalmente necrosada. No grupo controle, ocorreu reparação pulpar e formação de ponte de dentina a qual se apresentava completa no último período de avaliaçäo (60 dias). Estes resultados sugeriram que o sistema Sbmp foi irritante näo permitindo a reparaçäo, nem a formaçäo de ponte de dentina em polpas de ratos mecanicamente expostas.

ABSTRACT

Foram utilizados 120 primeiros molares superiores de 60 ratos, com o objetivo de avaliar a reaçäo da polpa sob a açäo de pastas à base de hidróxido de cálcio. Os animais foram distribuídos em 3 grupos de 10 ratos, divididos em 5 períodos de 4 animais de 3, 7, 15, 30 e 45 dias. Concluiu-se que: 1 - todas as pastas caracterizam-se como irritantes, persistindo no decorrer dos períodos evoluçäo por reparaçäo; 2 - houve melhor comportamento nos grupos 1 (hidróxido de cálcio + polietileno glicol 400) e iii (hidróxido de cálcio + polietileno glicol 400 + óxido de zinco) sobre o II (hidróxido de cálcio + polietileno glicol 400 + iodofórmio); 3 - todas as pastas, a partir do 7- dia, induziram à formaçäo de massas amorfas como mineralizadas, tendendo a barreira, apresentando variaçöes na qualidade e na quantidade, acompanhando o comportamento reacional
